Ethchlorvynol is a member of the class of tertiary carbinols with sedative and hypnotic properties. Ethchlorvynol appears to depress the central nervous system by binding at a benzodiazepine–GABA-A-chloride ionophore receptor complex, located in the limbic, thalamic and hypothalamic regions of the central nervous system (CNS). This increases the frequency of chloride channel opening, allowing the flow of chloride ions into the neuron and ultimately leading to membrane hyperpolarization and a decrease in neuronal excitability.
Ethchlorvynol is a sedative and hypnotic drug. It has been used to treat insomnia, but has been largely superseded and is only offered where an intolerance or allergy to other drugs exists.
Mechanism of action
Although the exact mechanism of action is unknown, ethchlorvynol appears to depress the central nervous system in a manner similar to that of barbiturates. Barbiturates bind at a distinct binding site associated with a Cl– ionophore at the GABAA receptor, increasing the duration of time for which the Cl– ionophore is open. The post-synaptic inhibitory effect of GABA in the thalamus is, therefore, prolonged.
